Application of HPMC in pharmaceutical preparation

1. Used as film coating material and film forming material

Hydroxypropyl MethylCellulose HPMC is used as film coating material. Compared with sugar-coated tablets, the coated tablets have no obvious advantages in covering drug masking smell and appearance, but their hardness, friability, moisture absorption, disintegration, coating weight gain and other quality indexes are better. The low viscosity grade is used as water-soluble film coating material for tablets and pills, the high viscosity grade is used as film coating material for organic solvent system, and the concentration is usually 2.0%~20%.

2. As adhesive and disintegrating agent

The HPMC low viscosity grade can be used as the adhesive and disintegrating agent of tablets, pills and granules, while the high viscosity grade can only be used as the adhesive. The amount varies according to the model and requirements. Generally, the amount of adhesive used for dry granulation tablets is 5%, and that used for wet granulation tablets is 2%.

3. As suspending agent

Suspending agent is a kind of viscous gel material, hydrophilic performance, by the use of suspending agent can slow down the sedimentation of particles, and it can be attached to the particle surface, to prevent the particle polymerization from forming a group. Suspending agent plays an important role in the production of mixed-suspension. HPMC is an excellent variety of suspending agent, its dissolved colloidal solution can reduce the tension of liquid-solid interface and the free energy of solid small particles, and then enhance the stability of heterogeneous dispersion system. The high viscosity level of this product as suspension preparation of suspension type liquid preparation, suspension effect is good, easy to re-disperse, non-stick to the wall, fine flocculating particles, its usual dosage is 0.5% ~ 1.5%.

4. As a blocker, slow and controlled release agent and orifice agent

The high viscosity grade of this product is used for the preparation of hydrophilic gel sustained release skeleton tablets, mixed material sustained release skeleton tablets as a blocker and controlled release agent, and has the effect of delaying the release of drugs, and its use concentration is 10% ~ 80% (W/W). The low viscosity grade is used as a pore-causing agent in sustained-release or controlled-release preparations where the initial dose required for the therapeutic effect of such tablets can be rapidly achieved and then sustained or controlled-release action is happened, with the effective plasma concentration maintained in the body. HPMC hydrates to form gel layer when it meets water. The release mechanism of drugs from skeleton tablets is mainly the diffusion of gel layer and the dissolution of gel layer.

5. Protective gel for thickening agent and colloid

When this product is used as thickener, the usual concentration is 0.45%~1.0%. This product can also increase the stability of hydrophobic glue, form protective colloid, prevent particles coalesce, condensation, thus inhibit the formation of sedimentation, its common concentration is 0.5%~1.5%.

6. As capsule material

Gelatin is usually the main material of capsule shell. The production process of Gelatin capsule shell is simple, but there are some problems and phenomena, such as poor protection to wet and oxygen are sensitive drugs, lower dissolution degree of drugs, and delayed disintegration of the capsule shell in the storage process. So HPMC as a substitute of Gelatin capsule material for using in the preparation of capsules, improve the capsule manufacturing formability and use effect, has been widely promoted at home and abroad.

7. As biological adhesive

Biological adhesion technology, the application of excipients with biological adhesion of polymer, by adhering to the biological mucous membrane, enhance the continuity and tightness of the preparation and mucous membrane contact, so that the slow release of drugs, and absorption by mucous membrane, to achieve the purpose of treatment, now widely used in the treatment of nasal cavity, oral mucosa and other parts of the disease. Gastrointestinal biological adhesion technology, is a new kind of drug delivery system developed in recent years, it not only extend the retention time for the pharmaceutical preparations in the gastrointestinal tract, and improve the contact performance of drugs and cell membrane absorption , change the cell membrane fluidity, that the penetration of drugs on small intestinal epithelial cells, so as to improve the bioavailability of drugs.

8. Gel agent for local use

As a skin adhesive agent, gel agent has a series of advantages such as safety, aesthetics, easy cleaning, low cost, simple preparation process, and good compatibility with drugs. In recent years, it has been widely concerned and become the development direction of skin external preparation.
